- System include:
• The system must use partition-based digital PCR technology (e.g., droplets, wells, chambers, nanowells, or equivalent).
• The system must perform endpoint PCR detection with positive/negative partition classification.
• The system must calculate absolute target concentration using Poisson statistical analysis.
• The system must generate ≥ 20,000 accepted partitions per reaction.
• The system must report total partitions generated, accepted vs. rejected partitions, and partition acceptance criteria.
• The system must support DNA dPCR without requirement for external calibration curves.
• The system must support RNA analysis through one-step RT-dPCR or two-step RT followed by dPCR.
• The system must support commercially available RT-dPCR reagents.
• The system must provide thermal protocols suitable for reverse transcription and PCR amplification in the same or sequential workflows.
• The system must support ≥ 4 fluorescence detection channels.
• The system must provide spectral separation and crosstalk correction.
• The system must support duplex assays for internal controls and inhibition monitoring.
• The system must provide temperature accuracy of ± 0.25 °C or better.
• The system must provide temperature uniformity of ≤ ± 0.5 °C across reactions.
• The system must include evaporation control (heated lid or equivalent).
• The system software shall provide absolute quantification results with confidence intervals.
• The system shall report: positive and negative partition counts, accepted partition counts, and target concentration with uncertainty.
• The system shall export data in CSV/XLSX and PDF formats.
• The system shall support multiple user roles and audit logging.
• The system shall allow storage and retrieval of historical runs.
• The system shall support No Template Controls (NTCs).
• The system shall support positive controls.
• The system shall support inhibition controls.
• The system shall provide run acceptance criteria and automated flagging of failed controls.
